Method notes

Assumes volumes are additive and the diluent is compatible with the stock solvent. Check final solvent percentage (for example DMSO) before adding to cells.

How dilution math works

C1V1 = C2V2

The amount of solute does not change when you dilute — only the volume does. So stock concentration × stock volume equals final concentration × final volume.

Solving for stock volume

V1 = (C2 × V2) / C1

Then add diluent up to V2 — do not add V2 on top of V1.

Serial dilutions

A 10-fold series moves 1 part into 9 parts diluent at each step. Errors compound geometrically, so mix thoroughly and change tips between tubes.

When not to use this

Percent w/v and percent v/v solutions follow the same algebra, but mass-based dilutions of viscous or volatile solvents need density correction.
